ABSTRACT

AIM: To evaluate the pathogenic variants of the SCO2(OMIM 604272)gene in patients with high myopia from Enshi Tujia and Miao Autonomous Prefecture of China.METHODS: A total of 384 patients with high myopia whose spherical refractive error was ≤ -6.00 D and whose axial length was ≥26.00 mm in at least one eye were recruited. DNA was extracted by the phenol-chloroform method from 5 mL of peripheral venous blood. Sanger sequencing was performed to identify pathogenic variants in exon 2 of SCO2. The detected variants were evaluated via in silico prediction software. A total of 288 people from the same district were included as the normal control cohort.RESULTS: Seven variants were detected, namely, four synonymous variants(c.201C>T/p.=, c.576C>T/p.=, c.633A>C/p.=, c.780T>C/p.=.), two missense variants(c.187A>G/p.Ile63Val, c.59G>C/p.Arg20Pro)and one nonsense variant(c.544C>T/p.Gln182*). The two missense variants were not damaging, as predicted by PolyPhen2, SIFT and Provean. The novel nonsense variant(c.544C>T/p.Gln182*)cannot be found in the 1000 Genomes Project and was not identified in 288 normal controls. Variant Taster suggested that the nonsense variant site was conserved.CONCLUSION: The newly identified nonsense mutation may be responsible for high myopia of the patients in our cohort. SCO2 is associated with high myopia, while the incidence of SCO2 variants in high myopia in this cohort was as low as 1/384; the nonsense mutation may be a scarce variant of high myopia in the Enshi Tujia and Miao Autonomous Prefecture of China.

ABSTRACT

Optical coherence tomography (OCT) is an optical diagnostic technique that provides non-contact, non-invasive tomographic imaging of microscopic structures of living ocular tissues.However, it requires the patient to be seated during the examination, which limits its use in infants and recumbent patients.Intraoperative OCT (iOCT) can be used in patients in the decubitus position to provide the surgeon with critical real-time information on anatomy and subtle lesions, improving the success and safety of the procedure.At present, the clinically applied iOCT types include handheld OCT, microscope-mounted OCT, microscope-integrated OCT and so on, among which integrated microscope-integrated OCT is the most widely used.iOCT is mainly used in a variety of fundus surgeries, retinal detachment reduction and proliferative diabetic retinopathy resection in retinal disease surgeries, and macular hole repair and anterior macular membrane stripping in macular surgery.During surgery, the use of iOCT can play a role in reducing surgical complications, detecting subclinical lesions, helping the surgeon make the most appropriate surgical decision, and predicting patient prognosis.This article reviewed the classifications of iOCT, as well as its advantages and disadvantages in the application of retinal detachment, diabetic retinopathy, macular epiretinal membrane, macular hole and other diseases to provide a reference for clinical practitioners.

ABSTRACT

As the elderly age, the incidence of metabolic syndrome(MS)among them increases significantly.Meanwhile, metabolic disorders superimposed on aging-related degenerative organ changes lead to confusion over their pathogenesis and clinical manifestations and pose special considerations.This review discusses progress in research using the integrated traditional Chinese and Western medicine approach on the pathophysiological mechanisms and clinical symptomology and, from the perspective of dialectical treatment, examines the metabolic changes and mechanisms of MS, aiming to promote comprehensive health management of the elderly and to achieve early diagnosis and intervention of MS in this age group.

Objective@#To investigate the mutation of P4HA2 gene in Tujia high myopia patients.@*Methods@#Clinical data and genomic DNA were collected from 288 Tujia patients with high myopia, whose spherical error ≥-6.00 diopters and axial length≥26 mm.All coding exons regions of P4HA2 were screened in patients to detect causative mutation by Sanger sequencing.The detected mutation was further screened in 192 normal control chromosomes in the same district.The pathogenicity of genetic mutations was predicted through bioinformatics analysis.This study followed the Declaration of Helsinki.All patients or their guardians signed informed consent.@*Results@#Four variations of P4HA2 gene were found in 288 patients, including one missense mutations(c.145C>A), two in-containing mutations (c.1306-62C>T, c.82+ 22C>T) and one insertion mutation (c.179+ 16_179+ 17 ins T). Missense mutation c. 145C>A was predicted as suspicious pathogenic gene by Polyphen2.According to the standard of ACMG in the United States, the variation was uncertain in pathogenicity.@*Conclusions@#Missense mutation c. 145C>A in P4HA2 gene is a suspicious pathogenic gene mutation in Tujia patients with high myopia.

ABSTRACT

Objective :To explore difference of blood lipid levels among patients with retinal vein occlusion (RVO) from department of ophthalmology , those with hyperlipidemia from department of cardiology and healthy people .Methods :Blood lipid levels were compared among 35 RVO patients who visited department of ophthalmology for the first time ,35 patients with hyperlipidemia who visited department of cardiology for the first time and 35 healthy people undergoing physi—cal examination from Jun 2017 to Jan 2018. Results : (1) Compared with healthy group ,there were significant rise in blood levels of total cholesterol (TC) and triglyceride (TG) ,and significant reduction in blood level of high density lipoprotein cholesterol (HDL—C) in RVO group and hyperlipidemia group (P<0.05 or <0.01) ,and significant rise in blood level of low density lipoprotein cholesterol (LDL—C) in hyperlipidemia group , P= 0.001 ; (2 ) Compared with hyperlipidemia group ,there were significant reductions in levels of TC [ (5.73 ± 0.91) mmol/L vs.(5.09 ± 0.83) mmol/L] ,TG [ (2.31 ± 1.37) mmol/L vs.(1.81 ± 1.89) mmol/L] and LDL—C [ (3.11 ± 0.79) mmol/L vs.(2.55 ± 0.42) mmol/L] in RVO group , P<0.01 all.Conclusion : Blood lipid levels of RVO patients are significantly higher than those of healthy people , but those were reduced than hyperlipidemia group indicated that blood lipid levels rise is a pathogenic factor ,but not only one ,its other pathogenic factor should be investigate .

ABSTRACT

Objective To explore the effect of different doses of anthracycline on preoperative chemotherapy of breast cancer.Methods In 40 cases of breast cancer patients using small doses of anthracycline therapy, and classified as the control group, the other 40 patients using high-dose anthracycline therapy, and classified as observation group.Then observe two groups of patients were compared;the patients were in our hospital from January to December 2016 were treated.Results The total effective rate was 82.5% in the observation group and 62.5% in the control group, the difference between the two groups was statistically significant (P<0.05).The incidence of adverse reactions in the observation group was 0, the control group was 27.5%, and the incidence of adverse reactions in the two groups was statistically significant (P<0.05).Conclusion Different doses of anthracyclines on the clinical effect of chemotherapy before surgery for breast cancer research found that different the clinical effect of different doses produced by E100C and E75C, E100C can improve the treatment effect of the patients, and reduce the occurrence of toxicity, so it is worthy of reference.

ABSTRACT

Objective To screen small fragments with high antigenicity in order to overcome the defects of the full-length Marburg virus GP as a vaccine antigen.Methods Based on the structure and function of GP sequence [amino acids(aa) 1-681], three small fragments,including GP1△(aa 25-239), GPM(aa 250-520) and GP2△(aa 436-648), were expressed by prokaryotic cells and immunized into mice, and the serum specific antibodies were detected after different immunization time.The proliferation of spleen lymphocytes and the concentration of cytokines of immunized mice were also measured.Results ELISA test results showed that high humoral immunity of GP2△ was produced as the full-length GP group did, and was higher than that of GP1△ and GPM (P<0.05).GP2△ immunization groups exhibited a higher SI value in mouse splenic lymphocytes stimulated by ConA than the mixed GP immunization groups did(P<0.05), but the effect was the opposite when mouse splenic lymphocytes were stimulated by the mixed GP.In addition, the amount of cytokines IL-2 and IFN-γ of mouse splenic lymphocytes in the GP2△ group was larger than that of the saline group (P<0.05), but smaller than that of the mixed GP group.Conclusion The fragment GP2△ can induce not only the high humoral immunity as GP does,but also moderate cellular immunity, which can be used for vaccine design.

ABSTRACT

Objective: To establish a fast quantitative detection method for tartrazine, sunset yellow, orange Ⅱ sodium salt and allura red in safflower by solid phase extraction-electrospray ionization-high performance ion mobility spectrometry (SPE-ESI-HPIMS).Methods: The pigments were extracted by 70% ethanol with ultrasonic treatment, and then a polyamide SPE column was used to remove the complex matrix interference in safflower.The purified sample was then dissolved in 90% methanol and analyzed under the optimized IMS parameters.Results: The detection time of all the pigments was less than 20 ms.The limit of detection of tartrazine, sunset yellow, orange Ⅱ sodium salt and allura red was 0.17 , 0.15 , 0.30 and 0.25 μg·ml-1 , respectively.All the pigments showed excellent linearity within the range of 0.5-20 μg·ml-1 (r>0.990 0), the method recovery was 88.0%-98.9% , and the RSD was 1.5%-5.2% (n =6).Conclusion: The method is rapid, simple, highly sensitive and reproducible, and suitable for the rapid quantitative detection of illegal added staining substances in safflower.

Objective To study the effect of electroacupuncture on repair of spinal cord injury and its effect on somatosensory evoked potential ( SEP) in dog models of intervertebral disc prolapse. Methods Nine Beagle dogs were randomly divided into three groups. In the model group and electroacupuncture group, the dog disc prolapse models were made by balloon compression, and in the electroacupuncture group, electroacupuncture was used every day for 14 days after operation. The model group was not treated after surgery. Sham operation was performed in the control group. Each dog was scored according to the Texas Spinal Cord Injury Scale for Dogs (TSCIS) scores before surgery (day 0) and on days 1, 4, 7, 14 after surgery. At the same time, SEP wave was measured using an EMG Evoked Potential Measuring Systerm and its latency and amplitude were analyzed. Results There was a significant difference in TSCIS scores between the model group, electroacupuncture group and control group at 1 day after operation. There was a significant difference between the electroacupuncture and model groups at 14 days after surgery. The amplitude of SEP in the model and electroacupuncture groups was significantly different from that in the control group at 1 day after operation, and there was a significant differ-ence between the electroacupuncture and model groups at 14 days after operation. There was a significant difference in the latency of SEP between the model and electroacupuncture groups at 4 days after operation, and between the electroacupunc-ture and model groups after at 14 days after operation. Conclusions Electroacupuncture can effectively promote healing of spinal cord injury in dogs with intervertebral disc prolapse, improve the TSCIS scores, restore SEP waveform, shorten the latency and enhance the amplitude. SEP can reflect the degree of spinal cord injury to a certain extent, and can be used to evaluate the effect of electroacupuncture treatment in these dogs.

ABSTRACT

Late intraocular lens dislocation is one of the most severe late complications after phacoemulsification.It often occurs 3mo after the surgery.Different from early intraocular lens dislocation, late intraocular lens dislocation is caused by zonular dehiscence and contraction of the capsular bag many years after phacoemulsification.In recent years, the incidence of late intraocular lens dislocation gradually increases, having a risk of 0.1% after 10a and 1.7% after 25a.In the long-term follow-up patients who underwent cataract surgery, 90% had zonular insufficiency and capsular contraction.Among the multiple factors which may contribute to zonular weakness and capsular contraction, pseudoexfoliation is the most common cause, accounting for 50% of all the cases.Other risk factors include aging, high myopia, uveitis, trauma, previous vitreoretinal surgery, retinitis pigmentosa, diabetes mellitus, atopic dermatitis, previous acute angle-closure glaucoma attack, and connective tissue disorders.The understanding of these predisposing factors will suggest necessary preventions for high-risk patients in the future.

ABSTRACT

AIM:To study the type, degree and axial distribution of low vision astigmatism in preschool children.METHODS:A group of 3-6 years old children were selected for astigmatism screening, and statistical analysis was performed on the detected 445 eyes of 308 people.RESULTS:With more than 0.50D astigmatism criteria, astigmatism examination of 308 people, accounting for 36.2%, of which 137 eyes astigmatism, astigmatism 171 monocular.The five types of astigmatism were compound hyperopia 40.7%, mixed 35.5%, compound myopia 8.5%, myopia 8.3%, simple hyperopia astigmatism degree 7.0%;69.0% were mild, 16.6% moderate, 14.4% severe.Astigmatism axial distribution was with the rule for 54.9%, against the rule 28.8%, oblique 16.6%.In binocular astigmatism eyes, axial symmetry was in 35.8%, asymmetry in 64.2%.CONCLUSION:The main type of astigmatism in preschool children are compound hyperopia and mixed astigmatism.Astigmatism degree is mainly mild.With the increase of age, the detection rate of moderate and high astigmatism increased.

AIM:To investigate the application of retinal nerve fiber layer (RNFL) thickness detected by OCT in the diagnosis of primary open angle glaucoma (POAG) and its relationship with the mean defect of visual field.METHODS:A retrospective study was carried out.Totally 158 cases (158 eyes) of patients with POAG were selected as POAG group, including 81 cases in early stage, 47 cases in middle stage and 30 cases in late stage.At the same time, 50 cases of persons without eye related diseases were selected as the control group.The upper, lower, nasal and bitamporal RNFL thickness and mean defect of visual field were detected, and the patients with POAG were followed up for 3mo.RESULTS:The upper, lower, nasal and bitamporal RNFL in POAG group were thinner than those in the control, and the mean defect of visual field was higher than that in the control group at different time (P0.05).The RNFL thickness of various parts was higher than that in patients with middle and late POAG, and the mean defect of visual field was lower than that in patients with middle and late POAG (P0.05).Pearson correlation analysis showed that the upper, lower, nasal and bitamporal RNFL thickness was negatively correlated with the mean defect of visual field (r=-0.719,-0.615,-0.681,-0.518, P<0.05).CONCLUSION:OCT can monitor the change of RNFL thickness in patients with POAG, and has a negative correlation with mean defect of visual field, which can be used for early diagnosis of POAG.

ABSTRACT

This study investigated the development characteristics of Dermacentor everestianus under laboratory conditions. The time taken for D. everestianus to complete the whole life cycle was 110.2 days on average, and the average developmental durations of larvae and nymphs were 17.1 days and 29.5 days, respectively. The summation of the prefeeding, feeding, and preoviposition periods of females was 17.8 days, and the oviposition and egg incubation lasted for 18.1 days and 27.7 days, respectively. A highly positive correlation was observed between the weight of engorged female and the number of egg mass laid (r=0.947). The reproductive efficiency index and the reproductive fitness index were 7.1 and 6.1, respectively.

ABSTRACT

Marburg virus (MARV) is a member of the Filoviridae family and belongs to a non-segmented, single-strand and negative-sense RNA virus.Since the first discovery of virus in 1967, infections have broken out 14 times, causing the infection of 588 people and 482 deaths.The mortality is up to 82%.Marburg virus results in multiple organ infections , severe hemorrhagic fever and death .Currently, there are no available licensed vaccines or post-exposure treatment , but the vaccines have proved effective in experimental animals .This review briefly summarizes the structure , infection mechanism and the progress in vaccines of this virus .

OBJECTIVE@#To study the correlation between miR-21 and Treg/Th17 ratio in the microenvironment of rats with hepatocellular carcinoma.@*METHODS@#Diethylnitrosamine was used to build the hepatocellular carcinoma model of rats; the content of Treg cells and Th17 cells and the expression of miR-21 in the peripheral blood of rats with hepatocellular carcinoma were detected. The statistical analysis was performed on the correlation between miR-21 expression and Treg/Th17 ratio.@*RESULTS@#Hepatocellular carcinoma model of rats was successfully constructed. The proportion of Th17 cells among all CD4(+)T cells in the peripheral blood of rats with hepatocellular carcinoma was 5.319%, which was higher than the control group; while the proportion of Treg cells was 9.472%, which was higher than the control group. Treg/Th17 ratio in the model group was 1.781, compared with 1.478 in the control group. The expression of miR-21 was increased in the peripheral blood of rats with hepatocellular carcinoma and it showed a positive correlation with the ratio of Treg/Th17.@*CONCLUSIONS@#There is a positive correlation between the expression level of miR-21 and the ratio of Treg/Th17.

OBJECTIVE@#To discuss the abnormal expression of Wnt inhibitory factor (WIF1) in hepatocellular carcinoma cells and its regulating effect on the hepatocellular carcinoma invasion and metastasis factors of tissue inhibitor of matrix metalloproteinases-3 (TIMP-3) and caveolin-1.@*METHODS@#RT-PCR and Western blot were employed to detect the expression of WIF1 in six hepatocellular carcinoma cell lines of HepG2, Hep3B, Huh7, PLC/PRF/5, SMMC-7721 and MHCC97 and the immortalized human liver cell line THLE-3. Besides, Lipofectamine 2000 was employed to transfect the eukaryotic expression vector pcDNA3.1-WIF1 and blank plasmid pcDNA3.1 into hepatocellular carcinoma cell lines. Transwell assay was used to detect the effect of WIF1 on the invasion ability of hepatocellular carcinoma cells; Western blot was used to detect the effect of WIF1 on the expression of TIMP-3 and caveolin-1 in hepatocellular carcinoma cells, it also discussed the effect on the expression of β-catenin.@*RESULTS@#The expression of WIF1 in hepatocellular carcinoma cell lines was lower than that in the normal liver cell lines (P < 0.01); while there was basically no expression of WIF1 in the human highly metastatic cell line MHCC-97 and moderate expression in HepG2 and SMMC-7721. Therefore, HepG2 and SMMC-7721 were chosen as the further experimental cell lines. After transfecting the eukaryotic expression vector pcDNA3.1-WIF1 and blank plasmid pcDNA3.1 into hepatocellular carcinoma cell lines, compared with the blank plasmid group, the cell viability and invasion ability in the WIF1 group were all reduced (P < 0.01), the expression of TIMP-3, caveolin-1 and mRNA were all down-regulated (P < 0.01), and the expression of β-catenin was decreased (P < 0.01).@*CONCLUSIONS@#Because of down-regulation or missing of expression of WIF1 in hepatocellular carcinoma cell lines, the up-regulation of WIF1 expression can significantly inhibit the invasion and metastasis of HepG2 and SMMC-7721 of hepatocellular carcinoma cell lines, which are related to the up-regulated expression of TIMP-3 and down-regulated expression of caveolin-1 and may be realized through the Wnt/β-catenin signaling pathway.

Transwell assay was used to detect the effect of WIF1 on the invasion ability of hepatocellular carcinoma cells; Western blot was used to detect the effect of WIF1 on the expression of TIMP-3 and caveolin-1 in hepatocellular carcinoma cells, it also discussed the effect on the expression of β-catenin. Results: The expression of WIF1 in hepatocellular carcinoma cell lines was lower than that in the normal liver cell lines (P < 0.01); while there was basically no expression of WIF1 in the human highly metastatic cell line MHCC-97 and moderate expression in HepG2 and SMMC-7721. Therefore, HepG2 and SMMC-7721 were chosen as the further experimental cell lines. After transfecting the eukaryotic expression vector pcDNA3.1-WIF1 and blank plasmid pcDNA3.1 into hepatocellular carcinoma cell lines, compared with the blank plasmid group, the cell viability and invasion ability in the WIF1 group were all reduced (P < 0.01), the expression of TIMP-3, caveolin-1 and mRNA were all down-regulated (P < 0.01), and the expression of β-catenin was decreased (P < 0.01). Conclusions: Because of down-regulation or missing of expression of WIF1 in hepatocellular carcinoma cell lines, the up-regulation of WIF1 expression can significantly inhibit the invasion and metastasis of HepG2 and SMMC-7721 of hepatocellular carcinoma cell lines, which are related to the up-regulated expression of TIMP-3 and down-regulated expression of caveolin-1 and may be realized through the Wnt/β-catenin signaling pathway.
